Flies heterozygous for a hypomorphic allele of rpd3 have life-spans extended by 33% (males).

Other phenotypes: Flies heterozygous for a hypomorphic allele of rpd3 have life-spans extended by 52% (females) [pubmed]. Expression of Sir2 is increased 2-fold in rpd3 flies [pubmed]. Caloric restriction in flies results in decreased transcription of RPD3 [pubmed] and fails to further extend the life-span of rpd3 flies [pubmed]. Deletion of RPD3 extends life-span in yeast [pubmed] and increases SIR2-dependent rDNA silencing [pubmed]. Overexpression of SIR2 extends life-span in yeast [pubmed] and life-span extension by deletion of RPD3 is likely to be the result of increased SIR2 activity.
